| I. Background | |
| ITGA4 基因位于人類(lèi)染色體 2q31.3 區域，編碼整合素α4 亞基，屬于整合素α鏈家族。 該基因通過(guò)可變剪接產(chǎn)生多個(gè)轉錄本變體，與β1 或β7 亞基結合形成α4β1（VLA-4）或α4β7 整合素，介導細胞粘附、遷移及信號傳導。在免疫系統中，α4 整合素促進(jìn)淋巴細胞與血管 內皮細胞粘附，驅動(dòng)淋巴細胞遷移至炎癥部位或次級淋巴器官，對免疫應答和穩態(tài)維持至 關(guān)重要。 臨床研究顯示，ITGA4 與多發(fā)性硬化癥、克羅恩病、炎癥性腸病等疾病密切相關(guān)，是這些疾病的治療靶點(diǎn)。在腫瘤領(lǐng)域，其基因啟動(dòng)子高頻甲基化與結直腸癌、肺癌、慢性淋 巴細胞白血病等癌癥的發(fā)生發(fā)展相關(guān)聯(lián)，成為早期篩查的潛在生物標志物。 |
