| I. Background | |
| BAFF（B 細胞激活因子，B Cell Activating Factor），也稱(chēng)為 TNFSF13B（Tumor Necrosis Factor Superfamily Member 13B）或 BLyS（B Lymphocyte Stimulator），是一種屬 于腫瘤壞死因子（TNF）超家族的細胞因子。BAFF 主要由單核細胞、巨噬細胞、樹(shù)突狀 細胞和某些基質(zhì)細胞產(chǎn)生，在 B 細胞的存活、增殖和成熟中起關(guān)鍵作用。 BAFF 通過(guò)與受體結合，調節 B 細胞的發(fā)育和功能，其過(guò)度表達與自身免疫性疾病 （如系統性紅斑狼瘡、類(lèi)風(fēng)濕性關(guān)節炎）密切相關(guān)。BAFF 有三個(gè)主要受體：BAFF-R （BR3）、TACI 和 BCMA。BAFF 與 BAFF-R 結合后，激活 NF-κB 和 PI3K/AKT 通路，促 進(jìn) B 細胞的存活和增殖；TACI 介導 BAFF 和 APRIL（另一種 TNF 家族成員）的信號傳導， 調節 B 細胞的活化和抗體產(chǎn)生；BCMA 主要在漿細胞中表達，介導 BAFF 和 APRIL 的信 號傳導，支持漿細胞的存活。BAFF 的過(guò)度表達與系統性紅斑狼瘡（SLE）、類(lèi)風(fēng)濕性關(guān)節 炎（RA）、干燥綜合等自身免疫性疾病密切相關(guān)。 | |
